چکیده

Studies with deuterium as a tracer have shown that wheat germ and pig heart malic dehydrogenase act on the keto form of oxaloacetic acid (1, 2). The keto form of this acid has also been shown to arise in the enzymatic carboxylation of phosphorylenolpyruvate by wheat germ phosphoryl-enolpyruvate carboxylase (3) and by avian liver phosphoryl-enolpyruvate carboxylase kinase (2). In an attempt to extend further our studies on the mechanism of the stereospecific reaction catalyzed by aconitase (4), it was deemed necessary to synthesize enzymatically monodeuterocitrate from stereospecifically labeled monodeuteromalate prepared in turn by the action of fumarase (5,6). To determine the feasibility of this experimental approach a study was undertaken to establish whether the keto or enol form of oxaloacetic acid is utilized by the condensing enzyme in the formation of citrate from acetyl coenzyme A. The results obtained in this study demonstrate that the keto form is the substrate of the citrate condensing enzyme.

Mechanism-Based Studies of the Active Site-Directed Inhibition and Activation of Enzyme Transketolase

Derivatives of phenyl-keto butenoic acids have been reported to be inhibitors of pyruvate decarboxylase, (PDC). The inhibition of transketolase, a thiamine requiring enzyme such as PDF, by meta nitrophenyl derivative of 2-oxo-3-butenoic acid (MNPB) is reported here. These studies indicate that the inhibitor binds to the enzyme at the active site. A two-step inhibition was observed, first th...

Citritase, the Citrate-splitting Enzyme from Escherichia Coli

Two distinct and independent pathways for the breakdown of citric acid are now generally recognized to operate in bacteria. These include (a) the stepwise degradation of citrate via the Krebs cycle reactions (l-3) and (b) the breakdown of citrate into a Cz and Ck unit not involving CoAl (4-6). A third conceivable pathway of citrate breakdown would be via a reversal of the condensing enzyme (req...

The purification of citrate-condensing enzyme.

The citrate-condensing enzyme was isolated as a crystalline protein from pig heart by Ochoa et al. (1). These workers reported that pig heart contained considerable quantities of the condensing enzyme. We were surprised when we obtained much higher yields of this enzyme from pig heart tissue with a different extraction procedure (2). This observation led to a purification procedure that yields ...

Radiometric assay of citrate condensing enzyme (citrate synthetase) in liver tissue and mitochondria.

Citrate synthetase activity was determined in samples of commercially available citrate condensing enzyme. We measured the amount of radioactive citrate formed from the condensation of carbon-14 labeled acetyl CoA and oxaloacetate. The labeled citrate was oxidized by oxidative bromination; 14C02 evolved was collected and represented the amount of citrate synthesized. Enzyme activity as low as 1...

Spectroscopic Studies on Titanium Ion Binding to the Apolactoferrin

Titanium (Ti) is a relatively abundant element that has found growing applications in medical science and recently some of Ti compounds are introduced as anticancer drugs. In spite of very limited data which exist on the Ti metabolism, some proteins might be involved in the mechanism of action of Ti. Up to our knowledge, there is not any report in the literature concerning binding of Ti to apol...
